Book Description

019963551X 978-0199635511 July 31, 1997
This is a collection of affinity-based purification protocols with an international authorship from across the life sciences. The introductory chapter outlines recent advances in ligand-coupling, matrix choice, and operating modes. The second chapter describes methods for obtaining quantitative data from affinity processes. There are specific chapters describing methods for the affinity separation of proteins, nucleic acids, and oligosaccharides. Methods for immunoglobulin purification and immunoaffinity protocols are discussed in a further chapter, and the use of affinity tags in the recovery of engineered proteins in another. Finally, modern techniques for cell separations are outlined. The book presents detailed laboratory protocols together with troubleshooting advice and reviews of recent developments. This book is aimed at bench scientists, both those in analytical research laboratories, and those non-specialists looking to apply affinity methods in their work; postgraduate and post-doctoral researchers, and academic staff in analytical research and general bioscience laboratories.

Editorial Reviews

Review

"While the editor suggests that it is not intended as a textbook, this book would certainly provide a valuable additional source for courses on bioseparations. However, its most welcome reception will come from laboratory scientists at all levels..."--Journal of Medicinal Chemistry

"This publication is designed to provide novice laboratory technicians and researchers with the background and examples necessary to develop and apply an affinity purification scheme. Built around practical examples, the chapters provide step-by-step protocols for the affinity separation of proteins, nucleic acids, oligosaccharides, immunoglobulin purification, and immunoaffinity applications. The eight individually authored chapters are further enhanced with background and troubleshooting information as well as charts, graphs, example results, and references." -- Biosis, Vol 50, Issue 10, Oct 22, 1998

Since the innovative work of Porath et al. (1) in the 1960s, and the seminal texts of Lowe and Dean (2) and Dean et al. (3), the technologies of affinity separation have become widely accepted-perhaps now to the extent of being taken for granted. Read the first page
