The aim of this book is to describe and discuss the preparation of immuno-gold probes, the progress and development of preparation methods suitable for immuno-electron microscopy, and the use of immuno-gold labeling at the microscopic level. The perspectives and limitations of the different methods used in combination with immuno-gold are evaluated not only in the methodology chapters, but also in the chapters where the validity of immuno-gold labeling is exemplified in a successful application to biological questions. In addition to examining cryosections as one of the most common preparation methods for immuno-electron microscopy, an alternative embedding method, such as freeze substitution of rapidly frozen samples, and replica methods, such as freeze-etching, fracture label, label fracture, and whole mount, are discussed in detail.
