ABOUT THE MANUAL

This manual has been written for the Introductory Microbiology student enrolled in either a general or an allied health program. It is intended for those students who take microbiology during their freshman or sophomore year and who, typically, have had only one year (or less) of chemistry. Its aim is to familiarize the student with the basic procedures and equipment used in the microbiology laboratoryincluding—diagnostic microbiology, clinical sample and sampling, serological procedures, antibiotic sensitivity testing and so on.

The manual has several important features. Each exercise has been written to be reasonably self-contained. It features explanations of the salient points being demonstrated or tested. These explanations are meant to supplement and complement, not replace, the assigned text. The exercises have been prepared in a manner that will provide flexibility in their application, dependent on the time frame of the laboratory. This will also enable the exercise to be divided up for performance by individual members of the laboratory group.

DOING YOUR PART

Each exercise is designed to teach a new technique or illustrate a principle of microbial growth. The exercises are arranged in the -following sequence:

  • Background material discussing the theory and experimental design underlying the procedures used in the exercise;
  • Objectives of the laboratory exercise; Materials needed for the laboratory;
  • Procedures outlining the specific instructions for completing the laboratory exercise; and
  • Laboratory Report Form.

As you proceed through the manual, you may notice that the laboratory exercises get progressively more complex and the instructions get somewhat less comprehensive. You are expected to become more competent with each exercise, and you should be able to problem-solve some of the needed procedures for yourself.

Laboratory reports are required for all exercises, and report forms are included for each exercise. They require tabulation of the data acquired in the exercise and include questions based on the experimental procedures and principles demonstrated in the exercise. Many of the questions will require your application of the observed procedures or results to other situations.
