Non-selective inhibitors of cyclic nucleotide phospho-diesterase (PDE), such as theophylline, have been in clinical use since the last century and have been used extensively in research since the identification of PDE in 1958.
Various PDE isoenzyme families have been defined, leading to the development of a second generation of PDE inhibitors with selectivity for certain families. Some of these new substances are under investigation as potential anti-inflammatory drugs.
