Can you use both the USB 2.0 and the 3.0 port at the same time? Wondering if you can have different PCs plugged into both of these ports at the same time? Anyone tried that or know if that works?

Good thought, but no. First, there's only one port, and second, USB is not designed to interface a client device with multiple host computers. A NAS server with USB ports would allow you to connect it to multiple computers.
