I have a Tamron 18-270 lense on my 60D. It just fits in the case. I don't do it up as it looks like it will wear a hole in it at the flash shoe. For me it works perfect as I carry the camera on my bike in a tank bag. So it acts like a protective cover only.

No, the shoulder strap for your camera exits on each side of the flap to enable you to carry your camera. Much safer alternative to having a strap on the case.

Most likely, yes it will. I have a Nikon D3200 with a 50-200 mm lens and it's still a little spacey when I put it in the case.
